摘    要:以东海穞稻(Oryza sativa L.)为材料,研究了试管苗在最低量生长条件下长期保存的适宜条件。试管苗在含BA0.5mg/L和ABA5mg/L的培养基上,不经转移,保存了一年,移栽入土后生长基本正常。剑叶的酯酶同功酶酶谱不变,但多数农艺性状不如实生植株。种子后代与对照无显著差异。应用本方法,已成功地将普通野生稻(Oryza sativa L. F. Spomtanea,Roschev)的腋芽在试管中保存了13个月,并移栽成活。讨论了影响试管苗长期保存的若干因素和长期保存后判断种质稳定性的指标。

Abstract:The appropriate condition to long term storage of rice germplasm in vitro studied. It was found that rice seedlings (Oryza sativa L. cv. Donghai Ludao) could be stored on N6 or He-5 medium added abscisie acid (ABA) 5 mg/l and benzyl adenine (BA) 0.5mg/l for one year without intervallic subculture. After being transfered to soil, these plants grew normolly in the main. The esterase zymogram of flag leaf with (?)soelectric focusing indicated no obvious differences between stored plants and the control There were no significant difference between the plants of their self-crossed gene. rations and neither in quantitative nor in qualitative traits. The axillary buds of a wild rice (O. sativa. L. f. apontanea) and seedlings of a traditional cultivar were successfully stored in vitro for 13 months and survived after being transfered to soil. The plants kept the characteristics of their parents respectively.Some factors influenced long term storage in vitro such as tempereture, contamination caused by acarids were discussed and the methods for judging the genetic stability of plants after long term storage were suggested.
