dapA基因缺失对大肠杆菌L-苏氨酸产量的影响

摘    要:采用Red重组技术,将大肠杆菌K12的dapA基因置换,构建dapA基因缺失的突变株,通过发酵测定突变株L-苏氨酸的产量.获得的dapA基因缺失突变株命名为K12△dapA.结果表明:在相同培养条件下发酵48 h,重组菌株K12△dapA发酵液中积累的L-苏氨酸达3.71 g/L,是Ecoli K12原始菌株的3倍.d...

Abstract:Using Red recombination technique,the dapA gene in Escherichia coli 'K12'was replaced,then a mutant without dapA gene was gained,and it was named as K12△dapA.Thus the K12△dapA and Escherichia coli 'K12'were both fermentated under same condition for 48 h,the L-threonine content in K12△dapA broth was 3.71 g/L,which was 3 times of that in Escherichia coli 'K12'broth.The results indicated that dapA gene deletion improved the accumulation of L-threonine.
