植物基因工程及其在林木害虫防治上的应用

摘    要:植物基因工程主要包括目的基因的分离、基因工程载体的构建、植物细胞的遗传转化、转化细胞的组织培养和植株再生、外源基因表达的检测等几个方面。利用该技术目前已转化了20多种林木树种,并成功地将抗虫的苏云金芽杆菌内毒素基因和蛋白酶抑制剂基因转入林木树种。

Abstract:This paper reviews five parts about plant genetic engineering which are,respec-tively,the isolation of target gene,the construction of engineering vectors,the transforma-tion of plant cells,the tissue culture of transformed cells and transgenic plants regeneration,and the detection of foreign gene.In recent years,plant genetic engineering in forest treeshas made rapid progress and great achievements among insect resistance,disease resistanceand herbicide resistance of plants.According to this technique,Populus alba×P.grandi-dentata was successfuly transformed and obtained the first transgenic plant in 1987.Bynow,over twenty forest trees were transformed.Nowadays,Both the toxin gene fromBacillus thuringiensis and proteinase inhibitor gene from other plants were transferred intoforest trees,especially poplar because of its wide plantation severely damaged by insects.The utilization of these engineered trees should be environmentally safer than many trad-itional methods,and mainly because these trees affect only species that feed on them,andeven will generally be harmful to only a limited number of insect taxa.There exist someproblems such as insects counter-evolve to overcome their resistance,so measurementsshould be taken to solve the counter-volution.
