核桃楸组培快繁技术研究

摘    要:以核桃楸幼茎为外植体,分析在不同培养阶段不同培养基配方对其组培能力的影响。结果表明:核桃楸茎段在DKW培养基中培养7 d后,萌发率达到95%,芽体平均长度达到4.5 cm;增殖培养以培养基组合(DKW+1.0 mg/L 6-BA),增殖效果最佳,平均每个外植体出芽数为4.5个,新芽高度4.9 cm;最适生根培养基是添加了5.0 mg/L吲哚丁酸的DKW培养基,生根率可达20.1%。生根苗移栽至草炭︰蛭石︰沙子=1︰1︰1混合基质中,成活率96%以上。由此,成功建立了核桃楸组织培养快繁技术体系。

Technology of Tissue Culture and Rapid Propagation of Juglans mandshurica Maxim.

Abstract:Taking young stem of Juglans mandshurica Maxim. as explant,the influences of different culture stage and different medium formula on the ability of tissue culture have been analyzed. The results show,after culturing in DKW medium for 7 days,the germination rate of stem of Juglans mandshurica Maxim. reaches to 95%,the mean length of bud reaches to 4. 5 cm. The effect of multiplication is the best with the combination of DKW + 1. 0mg /L 6- BA. The mean buds for each explant are 4. 5,the height of new bud is 4. 9 cm. The most suitable medium for rooting is DKW added with 5. 0 mg IBA,with rooting rate reaching to 20. 1%. The survival rate of rooted seedling can be over 96% after transplanting into the mixed medium of peat,vermiculite and sand at the proportion of 1︰1︰1. The technological system of tissue culture and rapid propagation of Juglans mandshurica Maxim. has been established.
