酶法辅助水提马尾松花粉及其成分检测研究

摘    要:目的]研究α-淀粉酶和蛋白酶辅助水提马尾松(Pinus massoniana Lamb.)花粉的工艺参数。方法]以水提物得率为指标,在单因素试验基础上进一步通过正交试验优化了酶法辅助水提马尾松花粉的工艺,并进行了验证试验。结果]经过超临界CO2萃取技术脱脂后的马尾松花粉利用α-淀粉酶辅助水提,得到的优化工艺为:加酶量2.0%,料液比1∶16,提取温度75℃,提取时间3h,恒定pH值6.0。在优化后的α-淀粉酶水提松花粉提取物中添加蛋白酶进一步水提,得到的优化工艺为:加酶量1.5%,料液比1∶20,提取温度50℃,提取时间3h,恒定pH值7.0。在优化工艺参数下,水提物的得率为51.34%。结论]为更加合理和充分地利用松花粉资源提供科学依据。

Abstract:Objective] The research aimed to optimize the parameters of the water extraction of Pinus massoniana Lamb. pollen assisted by the α-amylase and the protease. Method] Accroding to the yield of the water extraction,the orthogonal test was used to optimize the technology of enzymatic assisted water extraction of Pinus massoniana Lamb. pollen based on the single-factor test. Result] The Pinus massoniana Lamb. pollen degreased by supercritical CO2 extraction technology was extracted by water assisted by the α-amylase,and the optimized technology was as follow:Adding α-amylase amount 2.0%,the ratio of material to water 1∶16,the extraction temperature 75 ℃,the extraction time 3 h,constant pH value 6.0. The Pinus massoniana Lamb. pollen extract extracted by water assisted by the α-amylase was extracted furtherly by adding protease,and the optimized technology was as follow:Adding protease 1.5%,the ratio of solid to liquid 1∶20,the extraction temperature 50 ℃,the extraction time 3 h,constant pH value 7.0. The yield of Pinus massoniana Lamb. Pollen by enzymatic assisted water extraction was 51.34% under the optimized process parameters. Conclusion] The study can provide the scientific basis for better reasonably and fully utilizing the Pine pollen resources.
