杜仲愈伤组织的诱导及植株再生的研究

摘    要:在附加不同浓度NAA,6—BA的MS培养基上对杜仲的下胚轴和子叶进行愈伤组织的诱导并获得两类不同的愈伤组织,一类是白色的结构松散的非胚性愈伤组织,虽然能继代培养,但在附加较高浓度的6─BA的MS培养基上都不能再生植株;另一类是黄绿色的结构紧密的有颗粒状突起的胚性愈伤组织.在附加适量的6—BA的MS培养基上能较容易再生小植株。实验表明,杜仲的下胚轴和子叶在附加1.0mg/LNAA和1.0mg/L6—BA的MS培养基上能100%诱导成胚性愈伤组织;胚性愈伤组织在附加0.5mg/LNAA和2.5mg/L6—BA的MS培养基上培养,植株再生频率高达92%;小植株转移到附加1.5mg/L的IBA的1/2MS培养基上,15d左右长出较粗的根。

Abstract:Two kinds of calli were induced from hypocotyl and cotyledon of Eucommia ulmoides Oliv.on MS media supplemented with different concentrations of NAA and 6 BA.One was no-embryonic calli with white color and loose structure,from which no embryogensis and plant regeneration were observed when it was cultured on MS media supplemented with high concentrations of 6 BA;the other was particulate embryonic calli with yellow green colour and compact structure,from which embryogenesis and plant regeneration were easily observed when this kind of calli was cultured on MS media supplemented with optimum concentrations of 6 BA.Hismlogical studies on calli corresponded with the above mentioned results.It was discovered that plant hormone combination and the concentrations of plant hormone were the deciding factors influencing embryonic callus and plant regeneration.Calli were induced 100 percent when explants were cultured on MS media supplemented with 1.0mg/L NAA and 1.0mg/L 6 BA.92 per cent of embryollic calli were regenerated into plants when cultured on MS media supple mented with 0.5ing/L NAA and 2.5ing/L 6 BA.Roots formed 15 days after the plantlets were transferred onto 1/2MS media supplemented with 1.5ing/L IBA.Studies on tissue culture of Euconmmia ulmoides Oliv.and its micropropagation and abstract of secondary metabolite are of great importance.
