转基因抗虫油菜的ELISA分析

摘    要:为选育高效抗虫油菜新品种,采用酶联免疫吸附法检测Bt杀虫蛋白基因在转基因油菜植株中的表达。结果表明,Bt杀虫蛋白基因在转基因抗虫油菜植株生长时期而不同发生变化。在第3-9叶期Bt杀虫蛋白基因稳定、高效地表达,Bt杀虫蛋白的表达量为6.68-10.52ng/mg(以鲜叶计),占植物可溶性蛋白的0.067%-0.105%。但从第11后期后Bt杀虫蛋白表达量显著地降低,只有0.28-2.00ng/mg,占植物可溶性蛋白的0.003%-0.020%。

Abstract:To select and breed a new variety of high yield and high resistance to insects,the expression of Bt insecticidal protein gene in the transgenic rapeseed plants was studied with ELISA.The results show that Bt insecticidal protein gene variedly expresses in the transgenic rapeseed plants with its growth stage.It stably and efficiently expresses during the 3rd~9th leaf stage;its expressing quantity is up to 6.68~10.52 ng/mg fresh leaf;its proportion to plant soluble protein is 0.067%~0.105%. But its expressing quantity apparently decreases after the 11th leaf stage to 0.28~2.00 ng/mg fresh leaf,and its proportion to plant soluble protein is 0.003%~0.020%.
