不同温湿指数下荷斯坦奶牛外周血抗氧化指标的变化及其与淋巴细胞凋亡的关系

摘    要:选择体重、胎次、泌乳量均接近的健康荷斯坦奶牛32头,研究不同温湿指数(THI)条件下奶牛生理常数、外周血淋巴细胞凋亡和细胞周期以及血液抗氧化指标的变化,探讨血液抗氧化指标与细胞凋亡的关系.结果表明:(1)夏季热应激时期奶牛的呼吸频率和直肠温度极显著高于冬季非热应激处理组(P<0.01).(2)热应激可明显地增加奶牛外周血淋巴细胞的凋亡率,淋巴细胞凋亡率在THI为83.58时达到最高,为17.79%,与非热应激处理组相比差异极显著(P<0.01).(3)在THI为78.09时,G0/G1期细胞比例下降到最低,S期和G2/M期细胞比例达到最高,与非热应激处理组相比较差异均极显著(P<0.01);同时外周血淋巴细胞的增殖指数在此时达到最大值.(4)奶牛外周血超氧化物岐化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)的活性在夏季高温季节比非热应激处理组均呈极显著下降(P<0.01);丙二醛(MDA)含量在夏季高温季节时呈极显著上升(P<0.01);抗活性氧(抗ROS)活性在THI为83.58时最低.(5)血液中SOD、GSH-Px和抗ROS的活性与奶牛淋巴细胞凋亡率均呈中等强度负相关(P<0.01);MDA含量与淋巴细胞凋亡率呈正相关(P<0.01).可见:(1)热应激显著提高了奶牛的直肠温度和呼吸频率;(2)热应激对奶牛血液抗氧化指标与淋巴细胞凋亡率的变化影响显著;(3)奶牛外周血抗氧化指标的变化与淋巴细胞凋亡存在显著的相关性.

Abstract:The relationship between the antioxidant index and lymphocyte apoptosis was studied by investigating physiological changes,peripheral-blood lymphocyte apoptosis,cell cycle and the antioxidant index of 32 dairy cattle,which had identical body weight,parities and milk yield under different temperature-humidity index(THI).The results showed that:(1) The rectal temperature and respiratory rate of dairy cattle under heat stress were significantly higher than those in winter(P<0.01).(2) When THI reached 83.58,peripheral-blood lymphocyte apoptosis rate reached 17.79%,indicating that heat stress significantly increased peripheral-blood lymphocyte apoptosis.(3) When THI reached 78.09,G0/G1 phase of cell cycle reached the lowest,while S and G2/M phase reached the highest separately(P<0.01).At the same time,the proliferation index reached the highest(P<0.01).(4) The activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX) and anti-reactive oxygen species(anti-ROS) of peripheral blood significantly reduced (P<0.01),while activity of malondialdehyde(MDA) significantly increased(P<0.01) when THI was 83.58.(5) The activities of SOD,GSH-Px and anti-ROS were negatively correlated with the rate of lymphocyte apoptosis(P<0.01),while the content of MDA was positively correlated with the rate of lymphocyte apoptosis(P<0.01).In conclusion:(1) Heat stress evokes the fluctuation of the rectal temperature and respiratory rate of dairy cattle;(2) Heat stress significantly affects the cell cycle,antioxidant index and peripheral-blood lymphocyte apoptosis;(3) There is significant correlation between the antioxidant index and peripheral-blood lymphocyte apoptosis.
