摘    要:以番茄Micro-Tom为材料,以GA信号转导因子GAMYB基因 SlMYB33为对象,利用GUS组织化学染色分析 SlMYB33在花和果实中的表达模式;其次,通过载体构建、遗传转化得到转基因植株,通过转基因植株的表型分析探究 SlMYB33对番茄开花及果实发育的影响。结果表明, SlMYB33主要在番茄雄蕊和柱头中表达;在番茄Micro-Tom中过表达 SlMYB33,获得OE3、OE4、OE5、OE6和OE7五个过表达株系;表型观察表明, SlMYB33的过量表达可导致番茄开花时间延迟,同时促进果实增大。

Abstract:In this study,takingtomato cultivar Micro-Tom as materialanda Gibberellin (GA)signal transduction factor SlMYB33as objective,wefirstanalyzed the expressionpattern of SlMYB33 in tomato flowers and fruitsby GUS staining,and then,the transgenetic plants were obtainedby vector construction and genetic transformation,after that,the effect of SlMYB33 on tomato flowering and fruit development was studied through phenotypic identification in the transgenic lines.The results indicated that SlMYB33was mainly expressed in the stamens and stigmas of tomato.Five SlMYB33-overexpression lines,namely,OE3,OE4,OE5,OE6 and OE7,were obtained.The phenotypic detection showed that overexpression of SlMYB33 delayed flowering and increased fruit size in tomato.
