蓬莱松组培无菌体系的建立

摘    要:为了建立蓬莱松组培无菌体系,比较了不同消毒时间、外植体类型、启动和继代培养基配方等对蓬莱松无菌体系建立的影响。结果表明,外植体宜选用蓬莱松幼嫩的茎段,采用0.1%升汞消毒8 -10min即可;启动培养基配方为MS + 6-BA 0.5mg/L + IBA1.0mg/L + 蔗糖30g/L + 琼脂4.5g/L,其腋芽诱导率可达60%。继代培养基最适配方为MS + 6-BA 0.6mg/L + IBA1.0mg/L + 蔗糖30g/L + 琼脂4.5g/L,启动培养基上诱导出的芽可在该培养基上正常生长,培养90天后,可形成丛生芽。组培无菌体系的建立可为后续蓬莱松组培快繁体系的完善提供前期基础。

Study on Establishment of Aseptic System of Asparagus retrofractus
Abstract:In order to establish the aseptic system of Asparagus retrofractus in tissue culture, the effects of different disinfection time, explant types, initiation and subculture medium formula on the establishment of aseptic system of A. retrofractus were compared. The results showed that the explants should be selected from tender stems of A. retrofractus, and sterilized with 0.1% mercuric chloride for 8-10 minutes; The initiation medium was MS + 6-BA 0.5mg/L + IBA1.0mg/L + sucrose 30g/L + agar 4.5g/L, and the induction rate of axillary buds could reach 60%. The most suitable subculture medium was MS + 6-BA 0.6mg/L + IBA1.0mg/L + sucrose 30g/L + agar 4.5g/L. The buds induced on the start-up medium could grow normally on this culture medium, and cluster buds could be formed after 90 days of culture. The establishment of aseptic system of A. retrofractus can provide a good foundation for the subsequent development of a complete tissue culture and rapid propagation system of A. retrofractus.
