摘    要:以水稻幼苗为材料,比较了3种不同DNA提取方法,获得了一种以CTAB法为基础的DNA的简便、快速提取方法,并对RAPD反应体系进行了优化。结果表明,改良CTAB法所提取的DNA产量高、质量好,完全能够满足RAPD分析的要求。通过对DNA模板、Mg2+、dNPT、Taq DNA聚合酶浓度等因素的试验,对水稻RAPD分析体系进行了优化。新建的分析体系扩增谱带清晰、多态性强,稳定性好。

Abstract:Three methods(CTAB method,SDS method and improved CTAB method) for the extraction of genomic DNA in rice seedling were compared.The result showed that the improved CTAB method was the best method for the extraction of DNA among three methods,and the DNA extracted by this method had higher yield,better quality,and could be used for RAPD-PCR reaction.A number of factors,which affected the results of RAPD-PCR reaction,such as quantity of template DNA,concentration of Mg2+,dNPT and Taq DNA polymerase,were studi...
