日粮中添加NCG对妊娠前期鄂尔多斯细毛羊子宫内膜层及胎儿发育的影响

摘    要:为了研究日粮中添加N-氨甲酰谷氨酸(NCG)对妊娠前期鄂尔多斯细毛羊子宫内膜层及胎儿发育的影响,试验选择受孕鄂尔多斯细毛羊母羊40只,随机分为3组:NCG1组饲喂基础日粮+0.30 g NCG/d(n=13),NCG2组饲喂基础日粮+0.40 g NCG/d(n=13),对照组饲喂基础日粮(n=14),开展妊娠期0 d至90 d的饲喂试验。结果表明:妊娠期第90 d,NCG1组胎儿肺脏重显著高于对照组,NCG2组胎儿肝脏重显著高于对照组(P<0.05);妊娠期第17天,NCG1组母羊子宫内膜OPN基因表达量显著高于NCG2组(P<0.05)和对照组(P<0.01),NCG2组母羊子宫内膜Integrinαvβ3基因表达量显著高于对照组(P<0.05);妊娠期第90 d,NCG2组和NCG1组胎儿肾脂肪BMP7(P<0.05)和UCP-1基因表达量显著高于对照组(P<0.01),而NCG1组(P<0.01)胎儿肾脂肪PGC-1α基因表达量显著低于对照组,NCG2组(P<0.01)胎儿肾脂肪PRDM16基因表达量显著低于对照组。在本试验条件下,妊娠前期日粮中添加NCG有效促进了鄂尔多斯细毛羊妊娠识别、早期胚胎定植和子宫内膜层发育,保障了胎儿早期程序化健康生长。

Abstract:This study investigated the effects of different NCG levels of diet on maternal endometrium and fetal development during pre-pregnancy of Ordos fine wool sheep.Forty healthy Ordos fine wool sheep during pregnancy were allocated at random into 3 groups:NCG1 group by the based diet+0.30 g NCG/d(n=13),NCG2 group by the based diet+0.40 g NCG/d(n=13),and control group by the based diet(n=14).The experiment period lasted for 90 days from 0 day to 90 days of gestation.The results showed that the fetal lung weight in NCG1 group and liver weight in NCG2 group were significantly increased compared to control group at 17 days of gestation(P<0.05);The gene expression level of OPN in maternal endometrium in NCG1 group was significantly higher than in NCG2 group(P<0.05)and control group(P<0.01),and the gene expression level of integrinαvβ3 in maternal endometrium in NCG2 group was significantly higher than in control group at 17 days of gestation(P<0.05);For NCG2 and NCG1 groups,a significant increase of BMP7(P<0.05)and UCP-1(P<0.01)expression level in fetal kidney fat were found in relation to control group,however,the gene expression level of PGC-1αin fetal kidney fat in NCG1 group(P<0.01)and NCG2 group(P>0.05)were lower compared to control group,and the gene expression level of PRDM16 in fetal kidney fat in NCG2 group(P<0.01)and NCG2 group(P>0.05)were lower compared to control group at 90 days of gestation.In the experimental conditions,the addition of NCG in diet during pre-pregnancy effectively promoted the pregnancy identification,early embryonic colonization and endometrial development of Ordos fine wool sheep,and ensured the early healthy fetal growth.
