摘    要:目的]制备恩诺沙星人工抗原及多克隆抗体。方法]采用活性酯法合成恩诺沙星的人工抗原,并通过紫外光谱扫描和SDS-PAGE电泳对其进行鉴定。利用免疫原(ENR-BSA)免疫新西兰大白兔,制备抗恩诺沙星的高亲和力和高特异性的多克隆抗体。结果]恩诺沙星成功地偶联到载体蛋白上。通过动物免疫,获得了高效价的抗血清,最高效价达到1∶100 000,说明人工抗原成功地诱导机体产生免疫应答。结论]恩诺沙星的人工抗原合成路线简单易行,可为进一步建立恩诺沙星的免疫检测方法奠定了基础。

Preparation of Artificial Antigen and Polyclonal Antibody of Enrofloxacin

Abstract:Objective] The research aimed to prepare the artificial antigens and polyclonal antibody of enrofloxain.Method] The artificial antigen of enrofloxacin was synthesized by using active ester method and it was identified by using UV wavelength scanning and SDS-PAGE.ENR-BSA was used to immune New Zealand white rabbits and prepare polyclonal antibody of enrofloxain with high affinity and high specificity.Result] Enrofloxacin was successfully coupled with carrier protein.High-titer antiserum was obtained after immunization and the highest titer reached 1∶100 000,which indicated that artificial antigens successfully induced the immune response.Conclusion] The synthesis route of ENR artificial antigen was simple and feasible.This research could lay the foundation for further establishment of the immune detection method of enrofloxacin.
