不同大麦品种成熟胚愈伤诱导与分化研究

摘    要:为了筛选不受生长季节限制的大麦遗传转化受体,分析、比较了我国15个大麦品种成熟胚在1种诱导培养基和3种分化培养基上的愈伤诱导和分化率。结果表明,大麦品种间成熟胚愈伤诱导率差异非常显著,早熟3号的诱导率高达98.7%,而扬农啤2号和鄂大麦9号则低于10.0%。成熟胚在形成愈伤组织过程中,伴随胚发芽现象,但愈伤诱导与胚发芽之间没有相关性;品种间愈伤组织分化率也差异显著,早熟3号的分化率为43.3%,而其它品种的分化率均在10.0%以下;不同分化培养基的分化效率不同,在M1培养基上的平均分化率较高。

Abstract:To screen barley varieties suitable for genetic transformation that is not constrained by seasonal restrictions in field,fifteen elite barley varieties from China were compared for their callus induction and differentiation derived from mature embryos on different media.The results indicated that there was a significant difference of callus induction frequencies in different varieties.Zaoshu 3 displayed a frequency of 98.7%,whereas Yangnongpi 2 and Edamai 9 had a frequency of less than 10.0% under the same conditions.Although mature embryos germinated during callus induction no significantly correlation was observed between germination and callus induction.Furthermore,differentiation frequencies among different varieties also varied significantly.Zaoshu 3 had a frequency of 43.3% and the frequencies for all the remaining varieties were less than 10.0%.Three differentiation media had different effects on differentiation of barley varieties,and M1 medium was better than M2 and M3.
