小叶锦鸡儿根瘤菌的分离及其16S rDNA PCR-RFLP分析

摘    要:对辽宁地区与小叶锦鸡儿共生的根瘤菌资源进行了初步调查。采自5个不同地区样品的根瘤,通过分离、纯化、回接验证等试验共获得65株供试根瘤菌菌株。进一步选用4种限制性内切酶对供试根瘤菌进行了16S rDNA PCR-RFLP研究,结果表明其系统发育地位位于中慢生根瘤菌属(Mesorhizobiumspp.),并在96%相似性水平上分为5个不同的类群,分别由相应的rDNA图谱组合代表。丰富度及频度分析表明,组合15是辽宁省的优势群,组合18丰富度居第二位,但频度最高,也是辽宁省的主要类群。

Abstract:The study aimed to investigate the rhizobia isolated from Caragana microphylla growing in Liaoning Province of China.The nodules from 5 different sites were separated,purified and cultured in YMA medium.The isolates'symbiotic ability was confirmed by nodulation test on their original host.Finally,65 isolates were obtained.Their 16S rDNA-PCR products,about 1.5 kb in size,were digested respectively with AluI,HinfI,HaeIII and MspI,and then separated in 3.0%(W/V) agarose gels.The RFLP patterns were combined in clustering analysis.The result showed that the phylogenic status of 65 isolates were in genera of Mesorhizobium,and at 96% similarity level.They could be classified into 5 different groups corresponding respectively with 5 rDNA types,which were further analyzed by richness and frequency.The group of rDNA type 15 with the highest richness was regarded as the dominant cluster in Liaoning.The rDNA type 18 group with the highest frequency and the second richness was also the main cluster in Liaoning.
