摘    要:采用稀释涂布平板法,从龙眼腐烂果实中分离细菌和真菌,并结合菌落形态观察以及16S rDNA和ITS序列分析对所分离的细菌和真菌进行鉴定。结果表明,采用稀释涂布平板法,从龙眼腐烂果实中分离得到11株细菌和7株真菌。菌落形态观察以及16S rDNA和ITS序列分析结果表明,所分离的细菌主要鉴定为奥斯陆莫拉菌、短小芽胞杆菌、柠檬明串珠菌、肠膜状明串珠菌和东方醋酸菌。所分离的真菌主要鉴定为淡色生赤壳菌、葡萄座腔菌科真菌、间座壳菌、疖葡萄座腔菌和木贼镰孢菌。从龙眼腐烂果实中分离到的细菌和真菌目前在龙眼采后病害中未见相关报道,对于其致病性检测还有待于进一步的回接验证,以明确导致采后龙眼果实腐烂的主导微生物。

Abstract:Bacterial strains and fungal strains were isolated from rotting Dimocarpus longan Lour. by method of dilution butteron on plates. Their colony morphology was observed and 16S rDNA and ITS sequences were analyzed. Eleven bacterial strains and seven fungal strains were isolated from rotting Dimocarpus longan Lour. by method of dilution butteron on plates. The results showed that, the bacterial strains were identified as Moraxella osloensis, Bacillus pumilus, Leuconostoc citreum, Leuconostoc mesenteroides and Acetobacter orientalis. Seven fungal strains were identified as Bionectria ochroleuca, Lasiodiplodia pseudotheobromae, Diaporthe phaseolorum, Botryosphaeria dothidea and Fusarium equiseti. These bacterial and fungal strains were never reported in postharvest diseases of longan. The further infecting tests would be conducted in order to make clear the dominant microbes.
