白菜型油菜小孢子培养成胚率及植株再生的影响因素

摘    要:以白菜型油菜为供试材料进行小孢子培养研究。通过对影响其成胚和成苗等因素的研究发现,产胚率受不同基因型的影响,差异较大;小孢子发育时期与花蕾形态具有一定的相关性,即花蕾大小为2.0~3.0 mm,80%以上小孢子处于单核晚期-双核早期,并且处在该时期的小孢子最易诱导成胚;换液培养和起始高温热激培养可明显提高产胚率;B5固体培养基中最佳琼脂质量浓度为12 g/L,琼脂质量浓度过低或过高很难诱导成苗。

Abstract:The microspore culture of Brassica campestris L.was studied in this paper.The results indicated that much great difference existed in the ratio of embryos among different genotypes.The certain relevance existed between the developing stage of microspore and bud size,more than 80% of the microspores were in the single nucleus later period to binuclear early time when the bud size was 2.0-3.0 mm,and the embryo was obtain easily at this stage.Embryo yield was enhanced through medium renovation and high tempera...
