荷花蜂花粉多糖提取条件的研究

摘    要:采用温浸法,在单因素试验基础上设计4因素3水平正交试验,确定荷花蜂花粉多糖最佳提取工艺条件,采用苯酚-硫酸法检测多糖含量。结果表明:4因素对荷花蜂花粉多糖提取的影响顺序为提取次数>提取温度>料液比>提取时间;荷花蜂花粉多糖提取的最优条件为1:8的料液比,50℃水浴条件下,浸提3h。考虑到经济效益和成本因素,经两次提取后荷花蜂花粉多糖的提取率就可以达到99%,故最佳提取次数为两次。

Abstract:In this paper,the influences of different extraction conditions like extraction times,extraction time,ratio of material to liquid and water bath temperature on the extraction rate of pollen polysaccharides of water lily were studied.A 4×3 orthogonal test based on the single-factor experiment was carried out to find the best extraction conditions.And the determination of polysaccharides in this article was by phenol-sulfuric acid method.The result showed that the extraction times played the greatest influence on the polysaccharides extraction rate,which followed by the ration of material to liquid and water bath temperature.Meanwhile,extraction time played the minimal one.The optimal extraction conditions was:the ratio between powder and water 1:10;the extraction temperature 50 ℃;the extraction time 3 h.The extraction percentage of pollen polysaccharides of water lily reached 99% after extracted twice.So,considering the cost and the economy-effectiveness,extraction twice should be the best.
