茯苓多糖的超高压萃取工艺优化及其在卷烟中的应用

摘    要:目的]研究超高压提取茯苓多糖的最佳工艺条件。方法]在单因素试验的基础上,采用正交试验法对茯苓多糖的超高压提取工艺进行优选,选用L9(34)进行正交试验,以茯苓叶多糖的得率和纯度为指标,考察超高压压力、pH、料液比和保压时间对茯苓多糖提取率和纯度的影响。结果]试验得出,茯苓多糖的最佳提取工艺条件为:压力250 MPa、pH 10、料液比1∶15 g/ml、保压时间12 min,此条件下茯苓多糖提取率为2.64%,纯度为24.61%。在卷烟中的初步应用显示,添加0.6%的茯苓多糖提取物后,能显著增加卷烟香气量和香气质,各香韵间的协调性较好,杂气降低,余味舒适,综合效果相对较好。结论]同传统方法相比,超高压提取方法得率高,提取时间短,是提取茯苓多糖的适宜方法,研究可为茯苓多糖的工业化生产提供科学依据。

Study on the Extracting Polysaccharides with UHP and Application in Tobacco

Abstract:Objective] To study the optimum conditions for extracting polysaccharides from Poria cocos with UHP.Method]On the basis of single factor experiment,using orthogonal experiment,the extraction technology for polysaccharides with UHP was optimized.Using L9(34) to do orthogonal experiment,taking the yield and purity of polysaccharides as indexes,the influence of UHP pressure,pH,solid / liquid ratio and pressure holding time to the extracting yield and purity of polysaccharides was investigated.Result] The optimum extracting conditions for polysaccharides are: UHP pressure of 250 MPa,pH of 10,solid / liquid ratio of 1∶15 g / ml,and pressure holding time of 12 min.Under the optimum extracting condition,the extracting yield of polysaccharides is 2.64%,while the purity is 24.61%.Conclusion] Compared with traditional extracting method,UHP is the best method in extracting polysaccharides,with higher extracting yield and shorter extracting time.The study will provide a scientific basis for industrial production of Poria cocos polysaccharides.
