摘    要:卵裂间隔(τ0)是与温度相关的参数,为早期胚胎发育时期第一次卵裂持续的时间,或连续两次细胞分裂的时间间隔,被广泛用于鱼卵染色体操作中最佳诱导时间的确定上。本文研究了史氏鲟♀×达氏鳇♂受精卵在14~22℃下卵裂间隔τ0与温度的关系,τ0为第一次卵裂(τⅠ)与第二次卵裂(τⅡ)间的时间间隔(τⅡ-τⅠ)。结果表明:在实验温度范围内,史氏鲟♀×达氏鳇♂受精卵τ0随着温度(T)的上升而减小,在14℃下τ0为85.0±1.3 min,22℃下τ0为45.8±2.2 min,两者之间符合线性回归关系:τ0=154.54-4.95T(R2=0.999 0)。22℃为史氏鲟♀×达氏鳇♂胚胎发育的温度上限,超过此温度,正常卵裂的胚胎比例将显著下降(<60%)。

Abstract:The mitotic interval(τ0) is a coefficient related with temperature and the first duration in the mitotic cycle during early synchronous embryonic cleavage or other intervals between two consecutive cell divisions.It has been extensively adopted to determine the optimum inducement time in chromosome manipulation of fish eggs.The relationship between mitotic interval of fertelized eggs and synchronous temperature under 14-22 ℃ for Acipenser schrenckii ♀×Huso dauricus ♂has been studied.In present paper τ0 was the first(2 cell stage,τⅠ) and second(4 cell stage,τⅡ) cleavage divisions(τⅡ-τⅠ) at various temperatures as 14,16,18,20 and 22(±0.1 ℃).It came to the conclusions that the mitotic intervals for Acipenser schrenckii ♀×Huso dauricus ♂ were 85.0±1.3 min at 14 ℃,76.1±1.5 min at 16 ℃,64.9±1.7 min at 18 ℃,55.6±1.8 min at 20 ℃,45.8±2.2 min at 22 ℃.There were strong,negative linear relationship between τ0(in minutes) and water temperature(T) for all five temperature application(τ0=-4.95T+154.54,R2=0.9990).The proportions of eggs undergone normal cleavage at 22 ℃ were drastically reduced(<60%) as compared with all other temperatures(85%-90%).It was indicated that 22 ℃ was the upper limit of water temperature tolerated by Acipenser schrenckii ♀×Huso dauricus ♂ embryos.
