白桦叶黄酮超声波提取及纯化工艺研究

摘    要:以白桦叶为原料,采用超声波法提取其黄酮化合物,以黄酮粗提物得率为指标筛选试验条件,进而进行响应面优化试验;采用NKA-9大孔吸附树脂对白桦叶黄酮粗提物进行纯化优化试验。结果表明:在料液比1∶41(g/g),乙醇质量分数90%,超声时间44 min的工艺条件下进行提取,黄酮得率可达6.12%。NKA-9大孔吸附树脂纯化粗提物的工艺条件为:上样浓度0.8 mg/mL,上样量3 BV,上样速率1.5 mL/min,pH 3,洗脱液乙醇体积分数90%,洗脱液用量2.5 BV。在此工艺条件下所得到的纯化物黄酮含量可达68.5%。

Abstract:The paper investigated the ultrasonic extraction of flavonoids from birch leaves (Betula platyphylla Suk.). The experimental conditions were optimized by response surface analysis with extraction rate of flavonoids as the indicator, and NKA-9 macroporous resin was used to purify the crude flavonoids extracts from birch leaves. The results indicated that, the optimal extraction conditions were: material-liquid ratio 1:41 (g/g), alcohol concentration 90%(m/m), and ultrasonic time 44 min, the yield of flavonoids was 6.12%. The technological conditions of NKA-9 macroporous resin purification were optimized as follows: sample loading concentration 0.8 mg/mL, sample volume 3 BV, flow rate 1.5 mL/min, pH 3, eluent alcohol concentration 90%, and eluent volume 2.5 BV, and the yield of purified flavonoids reached to 68.5% under these conditions.
