| 摘 要： | 选取40日龄依莎公鸡40只,随机分为2组,每组20只。对照组:每日腹腔注射生理盐水4 mL/kg,PB组:每日腹腔注射苯巴比妥80 mg/kg。连续处理5 d后,进行体外肝微粒体酶活性测定和体内探针药物安替比林(AP)消除动力学试验。结果表明,PB组的肝脏指数、肝微粒体蛋白含量和细胞色素P450含量、氨基比林-N-脱甲基酶(AND)、红霉素-N-脱甲基酶(ERND)和苯胺羟化酶(AH)的活性均极显著高于对照组(P<0.01),安替比林消除速率明显快于对照组。苯巴比妥对鸡肝微粒体CYP450具有明显诱导作用。

| Abstract: | Forty 40-day-old cocks were randomly divided into control group(physiological saline,4 mL/kg,i.p.× 5 d) and PB-treated group(phenobarbital,80 mg/kg,i.p.× 5 d),and the effect of phenobarbital on the hepatic cytochrome P450 in chickens was investigated by enzyme assays in vitro and antipyrine pharmacokinetics in vivo.In comparison with the control group,the concentration of microsomal protein,total content of cytochrome P450,activity of AND,ERND and AH,and metabolic rate of antipyrine increased significantly in PB-treated group.The results showed that hepatic microsomal cytochrome P450 enzyme in chickens could be obviously induced by phenobarbital. |
