金银花的过氧化物酶和淀粉酶同工酶遗传多样性研究

摘    要:目的]探讨不同产区金银花的亲缘关系。方法]采用聚丙烯酰胺凝胶垂直平板电泳技术,利用过氧化物(POD)同工酶和淀粉酶(AS)同工酶对10个地区11个金银花样品的遗传多样性进行分析。结果]金银花过氧化物酶同工酶谱带形成2个电泳区域,共有3个酶位点,Rf值分别为0.324、0.671和0.863。其淀粉酶同工酶谱也形成2个电泳区域,共有4个酶位点,Rf值分别为0.125、0.306、0.67和0.792。不同产区金银花的过氧化物酶和淀粉酶具有较丰富的多态性,说明金银花不同居群在遗传本质上是有区别的。聚类分析表明10个产区的11个金银花样品可聚为3个类群。结论]不同产区金银花的亲缘关系与其遗传本质和地理位置有关。

Abstract:Objective] The aim of the research was to discuss the genetic relationship of Lonicera japonica Thunb.in different producing areas.Method] By vertical polyacrylamide gel electrophoresis technology,the genetic diversity of 11 samples of L.japonica from 10 areas with peroxidase(POD) isoenzyme and amylase(AS) isoenzyme were analyzed.Result] The bands of POD isoenzyme in L.japonica formed 2 electrophoresis regions and had 3 enzyme sites with Rf of 0.324,0.671 and 0.863 resp.Its bands of AS isoenzyme also formed 2 electrophoresis regions and had 4 enzyme sites with Rf of 0.125,0.306,0.67 and 0.792 resp.POD and AS in L.japonica from different producing areas had more abundant polymorphism,which indicated that different populations of L.japonica was distinguishing on the genetic essence.The clustering analysis showed that 11 samples of L.japonica from 10 areas could be clustered into 3 groups.Conclusion] The genetic relationship of Lonicera japonica Thunb.in different producing areas were related with its genetic essence and geographical position.
