香蕉黑星病菌黑色素合成关键酶小柱孢酮脱氢酶基因SCD的克隆与分析

摘    要:形成黑色素化的附着胞从植物体表面直接侵入寄主是香蕉黑星病菌主要的侵染方式.通过利用Genome Walking方法对香蕉黑星病菌黑色素生物合成关键酶编码基因之一的小柱孢酮脱氢酶基因SCD进行克隆, 得到其序列全长为1696bp.序列测定及软件分析结果表明,该片段包括完整的开放读码框(738bp),其中含有一个长29bp的内含子,编码203个氨基酸.该序列具有典型的启动子结构,在5'端调控序列中发现几个典型的真核生物顺式调控元件TATA motif、CAAT-motif、TGAC -motif、ACTGC -motif等.试验结果为深入研究该基因的功能和结构奠定基础,并为进一步防治该病害提供新的理论依据及手段.

Abstract:Melaninzation appressoria directly penetrated into host tissue from plant surface is a main infection way of banana freckle causal agent P.musarum.Scytalone dehydrogenase gene(SCD) is one of encoding gene of melanin biosynthesis of banana freckle causal agent,and it was cloned by Genome Walking method and a 1696bp of sequence length was obtained.The sequence fragment contained a complete open reading frame(738bp),and an intron(29bp) encoding 203 deduced amino acid.The sequence had a typical promoter structure and several typical eukaryotic cis-regulatory elements such as TATA-motif,CAAT-motif,TGAC-motif,ACTGC-motif,and so on in the 5' regulation region.The experimental results would provide basis for further studying the function and structure of banana SCD gene,and new theory basis and method for controlling banana freckle causal agent.
