摘    要:分别在稀释、离心猪精液中添加0.2、0.4、0.8、1.6 mmol/L丁羟基甲苯(Butylated hydroxytoluene,BHT),15℃保存,检验保存35、d后精子TMS、PMI和NAR(%)、测定MDA浓度;确定并选择最佳BHT浓度用于猪精液冷冻保存,检验解冻后TMS、PMI、NAR和Mt-MP(%)、测定MDA浓度。结果显示:BHT显著提高稀释精液、离心精液各项指标百分率(P0.05),且MDA浓度显著降低(P0.05),BHT最佳浓度分别为0.8、1.6 mmol/L;0.8 mmol/L BHT显著提高冷冻-解冻精液各项指标百分率(P0.05),且MDA浓度显著降低(P0.05)。结果表明,BHT能通过抑制精子质膜氧化损伤,提高猪精液保存效果。

Abstract:In this study, the protection of BHT supplementation on spermatozoa function was evaluated with 0.2,0.4,0.8 and 1.6 mmol/L BHT during preservation of diluted and centrifugal boar semen at 15℃ ,then the percentages of TMS,PMI and NAR were tested at 1,2,3 and 5 d of preservation and concentration of MDA was determined at 3 and 5 d of preservation, respectively. Optimal concentration of BHT was chosen and applied to the freezing media of semen cryopreservation in 0.5 mL straws,after freezing-thawing,the percentages of TMS,PMI, NAR and Mt-MP were tested and concentration of MDA was determined. Results showed that the percentage of TMS,PMI and NAR were significantly increased than that of control in diluted and centrifugal semen (P<0.05) ,the optimal concentra-tion of BHT was 0.8 mmol/L and 1.6 mmol/L,respectively. Moreover,after added 0.8 mmoL/L BHT to the freez-ing media, the percentages of TMS, PMI, NAR and Mt-MP were increased (P< 0.05), and the concentration of MDA was reduced significantly than those of control (P<0.05). In conclusion,addition of BHT to dilute extender or freezing media improved the quality of boar semen by inhibition of spermatic plasma membrane oxidation.
