| 摘 要： | 对2005年四川资阳脑膜炎病例猪链球菌2型分离株ZYH33溶血素编码蛋白中包含多个抗原表位的第230～593氨基酸残基区域的基因片段进行扩增并克隆.基因片段经酶切处理后插入表达载体pQE-30的BamH Ⅰ和Sal Ⅰ位点之阃,构建融合表达质粒.转化宿主菌TG1经IPTG诱导后融合基因得到了表达,用猪链球菌2型菌体抗血清对表达的融合蛋白进行免疫印迹试验,分析融合蛋白的免疫反应性.试验结果提示该溶血素蛋白第230～593氨基酸残基区域可作为猪链球菌的诊断抗原,为基因工程疫苗的研制奠定基础. |

| Abstract: | Streptococcus suis serotype 2 strain ZYH33 was isloted in 2005 from the case of human meningitis in Ziy-ang of Sichuan provience.In this study,the gene fragment encoding 230 to 593 amino acid resides of ZYH33 suily-sin was PCR amplified and inserted the expression vector pQE-30 between the restriction enzyme sites BamH Ⅰ and Sal Ⅰ.The resultant plasmid was transformed into E.coli TG1 host cell and induced with about 45 ku.The protein reacted positively to Streptococcus suis serotype 2 rabbit sera.This work demonstrated that the fragment of suilysin 230- 593 could be an effective antigen candidate for Streptococcus suis diagnosis and may be useful in the develop-ment of diagnostic reagents and genetic enineering vaccines. |
