蕙兰原生质体分离条件的研究

摘    要:用蕙兰无菌苗的幼叶和幼原球茎以及花蕾时的花瓣和花粉块为材料,研究了纤维素酶(商品名OnozukaR-10)浓度、渗透压稳定剂甘露醇浓度和酶解时间等因素对蕙兰原生质体分离的影响,结果表明:①花粉块和花瓣的分离条件为1.5%纤维素酶、0.3%果胶酶、11%甘露醇、酶解时间6.5h时,其原生质体最高得率分别为4.56×106和2.12×106个/gFW。②幼叶的分离条件为1.0%纤维素酶、0.3%果胶酶、13%甘露醇、酶解时间20h时,其原生质体最高得率为3.36×106个/gFW。③幼原球茎的分离条件为1.0%纤维素酶、0.3%果胶酶、11%甘露醇、酶解时间16h时,其原生质体最高得率为1.87×105个/gFW。此外,花粉块与叶片的原生质体溶液中杂质很少,有利于提纯;花瓣原生质体溶液中碎屑较多,且有少量的针晶;原球茎原生质体溶液中的淀粉粒与针晶很多,非常不易提纯。因此,花粉块与叶片为蕙兰原生质体分离的较好材料

Study on Protoplast Isolation of Cymbidium faberi
Abstract:Factors affecting protoplast isolation of Cymbidium faberi, including isolation material, isolation time, and concentrations of cellulase and mannitol, were studied. The results were as follows. 1. The highest yield of protoplasts from pollens was 4.56×10 6/g FW when the protoplasts were isolated for 6.5 hours in an enzymatic solution containing 1.5% cellulase, 0.3% pectinase, and 11% mannitol. But in the same conditions, the highest yield of protoplast from petal was 2.12×10 6/g FW. 2. The highest yield of protoplasts from young leaves was 3.36×10 6/g FW when the protoplasts were isolated for 20 hours in an enzymatic solution containing 1.0% cellulase, 0.3% pectinase, and 13% mannitol. 3. When the protoplasts were isolated from young protocorms for 16 hours in an enzymatic solution containing 1.0% cellulase, 0.3% pectinase, and 11% mannitol, the yield of protoplasts was 1.87×10 5/g FW. In addition, the protoplasts isolated from pollens and young leaves were easy to be purified due to little impurities in the solution, and the protoplasts from petals and protocorms were difficult to be purified because of high contents of fragments of raphides and starch pellets. So pollen and young leaf were better materials for protoplast isolation of Cymbidium faberi.
