红花檵木CHS基因的克隆与序列分析

摘    要:查尔酮合酶(chalcone synthase,CHS)是进入类黄酮和花色素苷次生代谢的第1个关键酶。根据植物查尔酮合成酶保守区序列设计引物,以红花檵木(Loropetalurn chinense var.rubrum)大叶红的嫩叶为材料,用RT-PCR方法,分离得到了一个查尔酮合成酶基因的cDNA(GenBank登录号为JQ609678),将该基因命名为LcvrCHS1。该序列长927bp,编码232个氨基酸残基。其核苷酸序列与GenBank已登录的同样来源的核桃、山茶属植物CHS序列同源性达83%,与其他科植物(绣球花、葡萄、桃、马铃薯、甘草、领春木属)CHS序列同源性也达到80%以上;其编码的氨基酸序列与山茶属、葡萄、鳄梨、洋梨、沙梨、映山红CHS基因编码的氨基酸序列同样具有高度同源性,同源性高达98%。

Abstract:

Chalcone synthase (Chalcone synthase, CHS) is the key enzyme that catalyzes the first step in flavonoids biosynthesis and anthocyanins secondary metabolites. To get chalcone synthase gene of efficient expression Loropetalurn chinense var.rubrum, the young leaves (Loropetalurn chinense var.rubrum) were used, using RT-PCR method, cloned and sequenced, the CHS gene to rename LcvrCHS1 (GenBank accession number JQ609678). The sequence is 927bp length, encoding 232 amino acid residues. Which has 85% sequence homology with Walnut and Camellia that has been logged in GenBank; with other genus plants (Hydrangea, Grapes, Peaches, Potatoes, Licorice, Euptelea genus) , CHS sequence homology is also more than 80%; with other plants (Camellia, Grapes, Avocados, Bartlett pear, Sand pear, Azalea),CHS sequence also having high homology, up to 98% homology.
