摘    要:利用反转录多聚酶链式反应(RT-PCR)和RACE的方法克隆了家兔Dmrt1全长cDNA基因.氨基酸序列分析表明,家兔Dmrt1与其他哺乳类Dmrt1基因的氨基酸序列同源性较高,而与鱼类、两栖类、鸟类等氨基酸序列的同源性较低.RT-PCR分析表明,Dmrt1基因只在家兔的精巢和卵巢中表达,而在脑、垂体、肺、心脏、肝脏、肠、脾脏、肾、肌肉等组织中均不表达,且精巢中的表达量高于卵巢.这一结果表明,Dmrt1可能参与了家兔的性别决定过程.

Abstract:A full-length cDNA fragment of the Dmrt1 in rabbit was amplified by RT-PCR and RACE method, using total RNA extracted from testis. The sequence analysis indicated that the deduced amino acid sequence of the cDNA shared high identity with other mammalians, while low with fishes, amphibians and avian species. The RT-PCR analysis revealed that Dmrt1 mRNA was only expressed in testis and ovary but not in brain, pituitary, lung, heart, liver, intestine, spleen, kidney and muscle.
