1株番鸭源奇异变形杆菌的分离鉴定及其毒力基因分析

摘    要:为查清引起滁州地区某养殖场雏番鸭死亡原因,本研究通过对分离细菌的鉴定和动物致病性试验,确定奇异变形杆菌是引起此次雏番鸭发病的病原体,并将分离的奇异变形杆菌命名为AHcz-1株.根据其16S rRNA基因序列,建立其遗传进化树;根据已报道的8个毒力基因ureC、zapA、mrpA、ucaA、rsbA、pmfA、atfA、atfC序列,设计引物进行PCR扩增,并对毒力基因进行序列分析.结果显示:AHcz-1株与日本株(AB932526.1)亲缘关系最近,同源性高达100%;毒力基因pmfA、atfA、ureC均未发生变异;AHcz-1株具有较强的致病性;致病菌对丁胺卡那高度敏感,对庆大霉素、恩诺沙星中度敏感,对其它17种抗生素不敏感,表明AHcz-1株具有多重耐药性.本研究为番鸭源奇异变形杆菌临床诊断、临床用药及遗传变异研究奠定了基础.

Abstract:In order to explore the etiology responsible young Muscovy duck deaths on a farm in Chuzhou,the isolated bacteria were confirmed and identified to be Proteus mirabilis(P.mirabilis)based on biochemical characteristics and 16S rRNA gene sequencing.In addition,the obtained strain AHcz-1 was tested for the presence of the virulence genes(ureC,zapA,mrpA,ucaA,rsbA,pmfA,atfA and atfC)by PCR method.The evolution tree analysis based on 16S rRNA gene sequences showed that the isolate AHcz-1 was closely related to the Japanese strain(AB932526.1)as the nucleotide homology was as high as 100%.Analysis of virulence gene sequences did not find mutations on genes pmfA,atfA and ureC.The animal experiment demonstrated that this isolate had strong pathogenicity in ducks.Furthermore,drug susceptibility testing revealed its high susceptible to amikacin,moderately susceptible to gentamicin and enrofloxacin,and resistant to 17 other antibiotics.This first report of isolation of P.mirabilis from Muscovy duck cases in Anhui province provided a reference for clinical diagnosis,drug screening and genetic variation.
