摘    要:针对羊肚菌品种,特别是产业化培植品种,建立快速准确的品种鉴定方法。通过对来源确切的梯棱羊肚菌样品及同属5种其他羊肚菌进行ITS测序,并结合网络数据库中的梯棱羊肚菌ITS序列进行分析,获得梯棱羊肚菌的特异性鉴别引物。进一步对分子鉴别实验条件进行考察,以确定最终实验条件。通过对梯棱羊肚菌及羊肚菌近缘品种的分析,发现只有梯棱羊肚菌呈现特征电泳条带。表明该研究设计的特异性引物和优化的实验方法,可用于梯棱羊肚菌菌种的快速鉴定及产品的真伪鉴别。

Abstract:It is important to establish a rapid and accurate method for the identification of species for the cultivation morels varieties.The ITS(internal transcribed spacer)sequences was obtained from all of Morchella importuna and 5 other species of morels collected from different location in China.Combined with reliable online-sequences for homology alignment,the specific-species primers of Morchella importuna were designed according to the variation sites.And the optimal experimental conditions were obtained by optimizing conditions such as annealing temperature,annealing time and extension time,etc.The developed PCR method was applied in all samples.Morchella importuna showed unique characteristic band,and other species of morels had no band.The results showed that the developed specific primer and PCR approach can be used for rapid identification of Morchella importuna.
