棉花GhCYSTM9基因的克隆及在非生物胁迫下的表达分析

摘    要:CYSTM是真核生物中广泛存在的一类小分子蛋白,其在细胞信号转导、逆境防御反应中发挥着非常重要的作用。为了研究CYSTM基因在棉花非生物胁迫响应中的功能,本研究利用RT-PCR方法从陆地棉中克隆了GhCYSTM9基因。该基因cDNA全长237 bp,编码78个氨基酸,所编码蛋白具有一个C端保守的富含半胱氨酸的TM螺旋和特异的N端胞质元件。多重比对发现,GhCYSTM9与拟南芥CYSTMs具有较高的相似度,其中与AtCYSTM9(NP566703.4,AT3G22240.1)序列一致性最高,达68.33%。荧光定量PCR分析其组织表达特性发现,GhCYSTM9基因在雄蕊、花瓣、根和花萼中显著高表达,在叶片、茎和雌蕊中表达量较低。进一步研究发现,GhCYSTM9基因的表达受到低温、干旱和盐胁迫的诱导,在胁迫处理下,其表达水平均呈现显著的上调表达。非生物胁迫下的基因表达分析结果显示,该基因在棉花非生物胁迫响应中有重要作用。本研究为进一步开展GhCYSTM9基因功能研究奠定了理论基础。

Abstract:CYSTM is a class of small molecular proteins that widely exists in eukaryotes.They play a very important role in cell signal transduction and adversity defense response.In order to explore the function of CYSTM in cotton abiotic stress response,GhCYSTM9 gene was cloned from upland cotton by RT-PCR.The cDNA of GhCYSTM9 was 237 bp in length and coded 78 amino acids.The encoded protein contained a conserved C-terminal cysteine-rich TM helix and a unique N-terminal cytoplasmic element.Multiple sequence alignment showed that GhCYSTM9 protein sequence was similar to CYSTMs in Arabidopsis,which with the highest identity of AtCYSTM9(NP566703.4,AT3G22240.1)up to 68.33%.Quantitative real-time PCR analysis showed that GhCYSTM9 gene had significantly high expression in stamens,petals,roots and calyx,and had lower expression level in leaves,stems and pistils.Further analysis revealed that the expression of GhCYSTM9 was induced by low temperature,drought and salt stresses.The levels of gene expression were significantly up-regulated under stresses.The expression pattern under abiotic stresses indicated that this gene might play an important role in cotton abiotic stress response.This study laid theoretical basis for further researches on the function of GhCYSTM9 gene.
