茎尖部位与激素对甘蔗‘福农39’和‘新台糖22’愈伤过程的影响

摘    要:以甘蔗品种‘福农39’和‘新台糖22’的茎尖为外植体,以MS为基本培养基,研究不同激素浓度和不同茎尖部位对愈伤组织的诱导与分化的效应。结果表明:添加3.0 mg/L 2,4-二氯苯氧乙酸(2,4-D)对甘蔗品种‘福农39’愈伤组织的诱导效果最佳。在此浓度下,‘福农39’和对照‘新台糖22’愈伤组织诱导率分别达到98.9%和97.8%;茎尖生长点基部(较粗的一端)以上0?3 cm的部位愈伤组织诱导效果最好,‘福农39’和对照‘新台糖22’该部位的愈伤组织诱导率分别达到了97.8%和88.9%;添加2.0 mg/L 6-苄氨基腺嘌呤(6-BA)对甘蔗品种‘福农39’愈伤组织的分化诱导效果最好。在此浓度下,‘福农39’和对照‘新台糖22’愈伤组织分化出苗率分别达到了57.4%和74.1%;茎尖生长点基部以上0?3 cm部位诱导的愈伤组织分化出苗率最高,‘福农39’和对照‘新台糖22’在该部位的愈伤组织分化出芽率分别达到了41.5%和54.8%。本研究为后续‘福农39’转基因研究及健康种苗生产提供帮助。

Abstract:The stem tip of sugarcane varieties ’Funong39’ and ’ROC22’ were used as explant and MS as basic medium to study the callus induction and differentiation effected by different hormone concentrations and stem tip parts. The results showed that when the concentration of 2,4-dichlorophenoxyacetic acid(2,4-D) in the basic MS medium was 3.0 mg/L, the callus induction rates of sugarcane varieties ’Funong39’ and control ’ROC22’ were 98.9%and 97.8%, respectively. When using 0~3 cm above the base of stem tip(thicker end) growing point as explant, the callus induction rate of ’Funong39’ and ’ROC22’ reached the max value, which were 97.8% and 88.9% respectively.Addition of 2.0 mg/L 6-benzylaminoadenine(6-BA) to callus differentiation of sugarcane variety ’Funong39’ was the best. At this concentration, the callus differentiation rates of ’Funong39’ and ’ROC22’ reached 57.4% and 74.1%,respectively. The differentiation rates of calli induced by 0~3 cm above the base of shoot tip growth point was the highest, and the callus differentiation rate of ’Funong39’ and ’ROC22’ reached 41.5% and 54.8%, respectively. This research can provide help for future transgenic research of ’Funong39’ and the production of healthy seedlings.
