内蒙古地区典型春小麦亲缘关系的SRAP分析

摘    要:为明确和廓清内蒙古典型种植春小麦品种的亲缘关系,本研究利用正交设计法创建春小麦SRAP反应体系,并对其遗传多样性进行系统研究。结果显示:春小麦20μL SRAP-PCR最佳反应体系为:DNA模板量25 ng、正反向引物0.3μmol/L、2×Taq Master Mix 7.0μL;17对引物扩增出190个条带,其中112个条带具有多态性,多态比率为59.00%,平均每对引物有6.59个多态性位点,引物多态性信息量(PIC)平均为0.63;有效等位基因数(Ne)、香浓信息指数(I)和Nei's遗传相似系数(H)分别为1.6010、0.7101和0.5124;聚类分析结果表明,在遗传相似系数为0.10处,将7个春小麦品种分为2大类;在0.15处,第一大类又分为2个亚类。研究表明,地理来源相同的春小麦品种之间遗传差异较小;种植区域的环境自然选择是影响供试典型春小麦亲缘关系的重要因素。本研究所建立的春小麦SRAP反应体系稳定可靠、重复性好,条带清晰且多态性好,可为后续春小麦遗传关系、优异基因挖掘等研究具有一定参考价值。

Abstract:In order to further understand that the genetic relationship of spring wheat varieties in Inner Mongolia region,this study were established a wheat SRAP reaction system by using orthogonal design and studied the genetic diversity of spring wheat varieties.The results showed that the optimum reaction system of 20μL SRAP-PCR in spring wheats was as follows:forward and reverse primers 0.3μmol/L,2×Taq Master Mix 7.0μL,DNA template 25 ng;the 190 bands amplified by 17 pairs of primers,the 112 bands were polymorphic with a polymorphic ratio of 59.00%.The average number of polymorphic loci per pair of primers was 6.59,and the average number of primer polymorphic information(PIC)was 0.63;The effective allele number(Ne),Shannon-information index(I)and Nei's genetic similarity coefficient(H)were 1.6010,0.7101 and 0.5124,respectively;Cluster analysis showed that at the genetic similarity coefficient of 0.10,seven wheat species were divided into two major groups,and at 0.15,the first group was divided into two subgroups.The results showed that there was little genetic difference among wheat cultivars from the same geographical origin,and the genetic relationship of 7 wheat cultivars tested can be affected by geography.The established SRAP reaction system of spring wheats was stable,reliable,reproducible,clear and polymorphic,which can provide an effective reference for further research on wheat genetic diversity,genetic relationship and effective gene resources.
