石榴等陆生植物Actin1基因密码子偏向性与进化分析

摘    要:肌动蛋白Actin1基因参与构成植物细胞骨架,功能域高度保守,在植物系统演化中具有特殊作用,是重要的管家基因。本研究选取石榴等63种具有重要经济价值或生态价值的陆生植物Actin1基因,分析其密码子使用偏好性和进化。研究结果表明:石榴Actin1基因有效密码子值为46.809,密码子使用偏性较弱。石榴Actin1基因同义密码子相对使用度大于1的密码子大部分偏向于使用以G/C结尾,其偏性主要是由于自然选择压力影响。石榴等陆生植物的Actin1基因偏向使用AAC、GAU、CAG、GAG密码子,这种偏向性在陆生植物之间属于趋同进化模式,亲缘关系相近的物种对Actin1基因密码子使用模式相似。由63种陆生植物的Actin1基因密码子偏性分析证实与物种密码子偏性与其进化及亲缘关系相关。本研究通过对石榴等陆生植物Actin1基因密码子偏向性与进化分析为Actin1基因家族的后续研究提供理论参考。

Abstract:Actin1 gene,bearing highly conserved domains in plants,serves for the cytoskeleton construction and has an unique role in plant phylogeny and evolution,and thus is an important house-keeping gene.The research analyzed the codon usage bias and evolution of Actin1 genes from pomegranate(Punica granatum)and other 63 landplants with important economic value or ecological value.The results showed that the effective number codons of pomegranate Actin1 were 46.809,indicated a weak codon usage bias.The codons with the synonymous codons usage values≥1 in pomegranate Actin1 mostly ended in G/C bases.The natural selection pressure affected the codon usage bias of Actin1 from pomegranate.The codon usage bias of Actin1 genes from 63 land plants converged into the same preferred codons,including AAC,GAU,CAG and GAG,belonged to convergent evolution and the codon usage bias of Actin1 related to the relationships of species.This study provides a theoretical reference for the follow-up study of Actin gene fa mily by the codon usage bias and evolution analyses of Actin1 genes from Punica granatum and other land plants.
