香蕉ATG8g过表达植株的获得及启动子元件分析

摘    要:为研究自噬相关基因ATG8g在香蕉中的潜在功能,以及在过表达植株中对植株形态发育的影响,本研究通过农杆菌转化法获得MaATG8g拟南芥过表达植株。结果发现,MaATG8g对植株的外观形态不产生影响。同时将MaATG8g启动子连入pGreenⅡ0800-LUC载体,并对MaATG8g启动子序列分析,发现该启动子片段含有生长素应答元件、脱落酸响应元件、抗氧化反应元件、低温胁迫相关元件、水杨酸响应元件、赤霉素应答元件和其他逆境响应相关的顺式作用元件,推测目的基因具有胁迫相关性,可以进一步研究MaATG8g胁迫相关的生物学功能。

Abstract:In order to study the potential function of autophagy related gene ATG8 g in banana and its influence on plant morphological development in overexpressed plants, the overexpressing plants of Arabidopsis MaATG8 g were obtained by Agrobacterium mediated transformation. The results showed that MaATG8 g had no effect on plant morphology. Construction of pGreenⅡ 0800-LUC vector and the promoter of MaATG8 g was completed. The analysis of MaATG8 g promoter sequence found that the promoter fragment contained the auxin response element,abscisic acid response elements, oxidation reaction of related components, salicylic acid, low temperature stress response element, gibberellin response element and other adversity response related cis element, speculating that genetic had correlation with stress. The consequence could be used to do further research related to abiotic stress.
