2019年上海地区H9N2禽流感病毒分子特征及演化分析

摘    要:为了解上海市活禽市场H9N2亚型禽流感病毒(AIV)分离株的遗传变异情况,本研究对2019年分离的4株H9N2 AIV的8个基因节段进行PCR扩增、克隆和测序,并对获得的8个基因序列进行同源性以及基因进化分析,对与病毒适应性增加的关键氨基酸位点进行了分析,并和目前我国使用的H9N2流感疫苗毒株HA上的抗原位点进行了比较。结果:4个分离株的HA基因仍然属于Y280/97,8个基因节段的重组模式属于G57;裂解位点均为PSRSSR/GLF,符合低致病性AIV的分子特征;这4株病毒存在多个与适应性增加相关的氨基酸突变。在已报道的33个抗原位点中,这4株病毒与我国目前使用的2种H9N2禽流感疫苗毒株(A/chicken/Shandong/6/96(6/96)和A/chicken/Shanghai/F/98(F/98))相比较,最大差异18个抗原位点。以上研究为H9N2亚型AIV的防控和疫苗研制提供了科学参考。

Abstract:To understand the genetic variation of the H9 N2 subtype avian influenza virus(AIV)in Shanghai live poultry markets,all the eight genes of the four isolates obtained in 2019 were amplified,cloned and sequenced.The homology and phylogenetic analysis of the genes were made.The amino acid substitutions in the four viruses associated with an increase in viral fitness were summarized and the antigenic sites of the four viruses were compared with the vaccines used in China.The results showed that the HA gene of these isolates belonged to Y280/97 and that the recombination of the eight segments clustered with G57.They had a common motif PSRSSR/GLF at the cleavage site which indicated that they were lentogenic.The four viruses possessed multiple amino acid substitutions associated with an increase in viral fitness.Of the 33 sites investigated,our isolates had a maximum of 18 amino acid differences,compared with the two vaccines used in China(A/chicken/Shandong/6/96(6/96)and A/chicken/Shanghai/F/98(F/98)).These data provided reference for prevention and control of H9 N2 subtype AIVs and for development of vaccines.
