| 摘 要： | 目的]探索山羊精液的冷冻方法。方法]以甘油为冷冻保护剂,采用细管法冷冻保存山羊精液,探讨甘油浓度和添加时间、冻结方法和解冻温度对山羊精液冷冻效果的影响。结果]在稀释液中添加6%和7%甘油的处理组中,冻后精子活率与复苏率均显著高于添加3%、5%甘油的处理组。配液时添加6%甘油的处理组中冻后精子活率与复苏率均显著低于在4℃平衡后添加的处理组。把平衡的山羊精液分别在离液氮面3 cm处熏蒸9 min和-80℃冰箱中冻结9 min,前者的精子复苏率显著低于后者。在解冻温度分别为40、45和50℃的处理组中精子冻后活率与复苏率显著高于解冻温度为55和60℃的处理组,说明解冻温度以40～50℃为宜。结论]在山羊精液冷冻保存中,甘油的最佳添加时间为4℃平衡后冻结前。

| Abstract: | Objective] The aim of the research was to explore better freezing method of goat semen.Method] With glycerol as the cryoprotective agent,goat semen was cryopreserved by using plastic tube method.And the effects of the concn.and adding time of glycerol,freezing method and thawing temperature on the freezing effect of goat semen were studied.Result] The sperm motility and recovery rate after freezing in the treatment groups of adding 6% and 7% glycerol to diluent were both significantly higher than that in the treatment groups of adding 3% and 5% glycerol.The sperm motility and recovery rate after freezing in the treatment group of adding 6% glycerol in the period of preparing the liquid were both significantly lower than that in the treatment group of adding glycerol after equilibrium at 4 ℃.The balanced semen was suffocated above 3 cm of the liquid nitrogen surface for 9 min and frozen in-80 ℃ refrigerator for 9 min resp.The sperm recovery rate of the former was significantly lower than that of the latter.The sperm motility and recovery rate after freezing in the treatment groups with the thawing temperature of 40,45 and 50 ℃ resp.were all significantly higher than that in the treatment groups with the thawing temperature of 55 and 60 ℃,which indicated that the optimum thawing temperature was 40-50 ℃.Conclusion] In the goat semen cryopreservation,the optimum adding time of glycerol was the time after equilibrium at 4 ℃ and before freezing. |
