摘    要:利用RAPD技术对菜薹人工老化种子DNA的损伤及PEG渗调引发处理的修复作用进行了研究。在61种引物中,有6种在菜薹人工老化的种子上获得了基因组DNA指纹图谱,平均每种引物扩增的总DNA带数为2.16,多态性DNA带数为1.17,有11种引物在PEG渗调引发的菜薹老化种子上获得了基因组DNA指纹图谱,平均每种引物扩增的总DNA带数为3.91,多态性DNA带数也是3.91。人工老化对菜薹种子基因组D

Abstract:In this paper,RAPD was applied to genomic DNA polymorphism analysis of artificial aged and PEG osmoprimed artificial aged seed of flowering chinese cabbage.Among 61 primers used,6 amplified genomic DNA fingerprints on artificial aged seeds,the average number of DNA bands amplified by each primer was 2.16,and number of polymorphic DNA bands was 1.17.11 amplified genomic DNA fingerprints on PEG osmoprimed artificial aged seeds,the average number of DNA bands amplified by each primer was 3.91,and number of polymorphic DNA bands was 3.91. Damage on genomic DNA of flowering Chinese cabbage by artificial accelerated ageing were obvious.With ageing, DNA bands amplified were weaker and weaker till they disappeared completely.Damaged genomic DNA of artificial aged seeds of flowering Chinese cabbage could be repaired through PEG osmopriming.Stronger and/or new DNA bands were amplified from PEG osmoprimed artificial aged seeds.
