小麦幼胚离体培养的研究初报

摘    要:【目的】筛选适宜小麦幼胚培养的培养基和转基因受体材料的优良基因型,为小麦转化、遗传等提供依据。【方法】以7个春小麦品种(系)的幼胚为材料,取开花后12~16 d的幼胚进行培养,20 d后统计幼胚一步成苗数和愈伤组织绿苗分化数。【结果】小麦幼胚在C17、MS和YP三种培养基上都能诱导一次成苗,其中YP培养基一次成苗率最高(75%),其次是MS(33.13%),最低的是C17(23.34%),YP培养基一次成苗的效果明显优于其它两种培养基,不同基因型间的愈伤组织绿苗分化率差异明显。【结论】筛选出适宜小麦幼胚培养一次成苗的YP培养基和一次成苗和愈伤组织分化绿苗率都较高的材料新春9号,此材料可作为幼胚培养和转基因受体材料的优良基因型。

Abstract:【Objective】The suitable culture medium of wheat immature embryos and good genotype of transgenic receptor materials were screened so as to offer some information to be used in genetic study and transformation in wheat.【Method】The immature embryo of seven cultivars of spring wheat which were grown for 12 to 16 days were cultured for 20 days and the number of nonrecurring seedling from immature imbryo and the number of differentiated green seedling from callus culture was counted.【Result】The wheat embryos could induce the nonrecurring seedlings when imbryos cultured in C17,Ms and YPculture medium.The seedling ratio was the highest when imbryos were cultured in YP culture medium(75%) as compared with that of the Ms culture medium(33.13%) and the C17 medium(23.34%) was the lowest,which indicated that the effect of nonrecurring seedlings cultured in YP culture medium were significantly better than that of the other two culture medium.There were significant differences of differentiation ratio of green seedling from callus culture between different genotype cultivars.【Conclusion】The seedling ratio and differentiation rate of green seedling from callus culture of Xinchun 9 was the highest which showed that the Xinchun 9 could be used as the best genotype for immature imbryo culture and transgenic receptor material.
