无患子总DNA提取方法研究

摘    要:针对无患子富含多酚、多糖、蛋白质及其他次生代谢物质的特点,以无患子叶片为材料,分别采用改良的CTAB法和CTAB与SDS相结合法提取不同种源无患子的总DNA。结果显示,改良的CTAB法提取的总DNA,得率较高,纯度好,可用于酶切、PCR扩增等后续试验,并能获得清晰、稳定的扩增产物。而采用CTAB与SDS相结合的方法得到的基因组DNA效果不稳定,无法检测出DNA。改良的CTAB法具有简单、经济的特点,可用于无患子总DNA的提取,成功地进行SRAP分子标记分析,为研究无患子遗传资源多样性奠定基础。

Abstract:In view of the large amounts of secondary metabolites such as polyphenol,polysaccharide and protein in leaves of Sapindus Mukorossi,the total DNA of different provenances were extracted by improved CTAB and CTAB combined with SDS methods.Results showed that the effect of the improved CTAB method was better,DNA yield was higher with high purity,could be used for the follow-up tests like enzyme cut,PCR amplification,could obtain clear and stable amplification product.Otherwise,the effect of CTAB combined with SDS method got genome DNA was not stable,DNA concentration was low,it always couldn't detect DNA.Improved CTAB method had a simple and economic characteristics,could be used for the Sapindus Mukorossi’s total DNA extraction,successful used to the SRAP molecular marker analysis.It’s laid the foundation for the research of Sapindus Mukorossi’s diversity of genetic resources.
