叶籽银杏种质遗传多样性的AFLP评价

摘    要:文章对来自日本和中国的8株叶籽银杏亲缘关系进行了评价,从64对EcoRI/MseI引物中筛选出7对清晰、多态性高的引物,对8个叶籽银杏种质的亲缘关系进行了AFLP分析。7对引物共产生841条谱带、240个特异位点和749条多态带,多态带的比例平均为89.06%,每对引物鉴别效率达到100%,所有种质之间相似系数(SC)在0.48~0.78之间,表明叶籽银杏单株问存有较大的遗传变异,中国的叶籽银杏具有较丰富的遗传变异。

Abstract:Genetic diversity of Ginkgo biloba var. epiphylla is assessed through AFLP in this pa- per. The genetic relationship of eight Ginkgo biloba var. epiphylla trees from Japan and China was analyzed in order to provide theoretical evidence for preservation and utilization of Ginkgo biloba var. epiphylla. By selecting seven clear and high polyrnorphic primer combinations from sixty-four EcoRI/MseI primer combinations, AFLP analysis on the genetic relationship of eight Ginkgo biloba var. epiphylla trees from Japan and China were conducted. These seven primer combinations pro- duced 240 specific loci and 841 bands, including 749 polymorphic bands. The percentage of poly- morphic bands was 89.06 %. The identification percentage per primer combination reached tol00 %. The genetic similarity coefficient among all materials varied from 0.47 to 0.78, which shows a large scale in genetic variation, The genetic variation of Ginkgo biloba var. epiphylla from China shows greater diversity than that from Japan.
