摘    要:将120只600日龄产蛋山麻鸭随机分为正常日粮组、1/2石粉组和1/4石粉组,自动生化分析仪测定更换饲料前1 d及更换饲料后1、3、5周血清钙、磷水平,观察缺钙日粮对蛋鸭血清钙、磷的影响.分离山麻鸭破骨细胞(OC),在此基础上添加不同浓度钙离子(2、3、5、7 mmol·L~(-1)),倒置显微镜观察体外培养OC形态,培养1、3、5、7 d进行抗酒石酸酸性磷酸酶染色(TRAP),7 d后扫描电镜观察象牙片吸收陷窝.结果表明:钙缺乏可导致蛋鸭血清钙、磷水平升高,降低钙磷比例.高浓度钙和较低浓度钙显著抑制OC生成及骨吸收活性.结果说明缺钙可诱导OC生成并发挥骨吸收功能,最终导致骨代谢性疾病.

Abstract:To investigate the effects of serum calcium and phosphorus on the model of calcium-deficient laying ducks, one hundred and twenty Shan sheldrakes were allocated into normal diet, half calcium carbonate and quarter calcium car-bonate groups randomly. Serum calcium and phosphorus were detected by automatic biochemistry analyzer one day before and one, three and five weeks after fed with above diets. Osteoelasts (OC) isolated from Shan sheldrakes were cultured with different concentrations of calcium(2,3,5,7 mmol·L~(-1)).OCs were identified by tartrate resistant acid phosphatase (TRAP) staining and observed by phase-contrast microscope at 1, 3, 5, 7 d after treated by calcium. 7 d later, OC activity was evaluated by assessment of bone slices absorption degree. The results showed that ducks' serum calcium, phosphorus increased, while the proportion of calcium and phosphorus decreased in groups with calcium-deficiency diets. OC formation and activity were inhibited by higher concentrations of calcium. These indicate that calcium deficiency can induce bone metabolic disease by stimulating OC formation and bone resorption.
