摘    要:目的]寻找高效、快速的薰衣草(Lavandula pedunculata)基因组DNA提取方法。方法]通过改良CTAB法提取薰衣草3个品种的基因组DNA。结果]经检测,所提样品OD260/OD280值在1.85左右,得率为454.50~1 093.35μg/ml,电泳条带清晰,无明显的拖尾现象;Eco RI酶切基因组DNA图谱表明,提取的DNA能被限制性内切酶完全酶切;以提取的DNA为模板,用1对随机引物进行RAPD-PCR,发现检测条带清晰,说明获得的DNA质量较高,可以满足相关的分子生物学研究需要。结论]该研究可以为薰衣草分子生物学的后续试验的开展奠定理论基础。

Extraction of L.pedunculata Genomic DNA Based on the Modified CTAB Method

Abstract:Objective] The aim was to find efficiency and rapid L.pedunculata genomic DNA extract method.Method] The genomic DNA samples of 3 L.pedunculata cultivars were isolated by modified CTAB method.Result] The results showed that the ratio of OD260/OD280 was about 1.85,yield was 454.50-1093.35 μg/ ml,electrophoresis bands was clear and had no significant tailing phenomenon.The fingerprint figure of DNA digested by restriction enzyme Eco RI showed that the extract DNA could be completely digested.Taking the extract DNA as template,the results of RAPD-PCR amplification by one random primer showed that the detected bands were clear.So the extracted DNA had a good quality for deeply molecular biology study.Conclusion] The study can lay theoretical basis for deeply carrying out the biological follow-on test of L.pedunculata.
