| 摘 要： | 从尼罗罗非鱼的细菌人工染色体基因文库中提取和纯化含有卵巢和脑芳香化酶基因的重组质粒DNA，通过简并PCR制备芳香化酶基因原位杂交探针，并用荧光素进行标记。结果显示，尼罗罗非鱼卵巢和脑芳香化酶基因位于2对不同的小染色体上，而不是位于性染色体上。结果提示：芳香化酶基因不是尼罗罗非鱼主要的性别决定基因。 |

| Abstract: | The recombinant plasmids containing ovary and brain aromatase gene from bacteria artificial chromosome(BAC)library of Nile tilapia,Oreochromis niloticus,were extracted and purified according to the instruction of QIAGEN Plasmid Mini Handbook with a little of modification.The probes of aromatase genes for the in situ hybridization were prepared from the plasmid DNA and labelled with biotin by degenerate oligo-nucleotide primed PCR(DOP-PCR).The result showed that the ovary and brain aromatase gene of Nile tilapia was located on two different pairs of small chromosome other than the sex chromosome.So the aromatase gene is not primary sex determining gene in Nile tilapia. |
