大口鲇与鲇及其杂种F1的蛋白质和同工酶电泳图谱的比较

摘    要:采用聚丙烯酰胺水平平板电泳法研究了大口鲇、鲇及其杂种F1的血清蛋白、肌肉蛋白以及肝、肾、脾、心、眼和肌肉等6种组织的醇脱氢酶(ADH)、乳酸脱氢酶(LDH)和酯酶(EST),分析了杂种F1及其与父母本的亲缘关系。结果表明,在LDH和ADH上,杂种F1与母本大口鲇相近。对比发现,肾脏和脾脏的EST同工酶电泳图谱可作为鉴别别大口鲇的生化遗传标记。

Abstract:This study used plane polyacrylamide gel electrophoresis,serum and muscle proteins and the polymorphism of three isozymes including alcoholdehydrogenase(ADH),lactate dehydrogenase(LDH) and esterase(EST) from six tissues,liver,kidney,spleen,heart,eye and muscle,of adult Silurus meridionalis,S.asotusand their hybrids were profiled and determined respectively.The relationship of S.meridionalis,S.asotusand their hybrid were studied.Comparison showedthat the hybrid was closer to the maternal parent,S.meridionalisthan to the other on LDH and ADH.They were monopolymorphic in the electrophorgramofprotein.In contrast,the specificities of species and tissues were detectedandthe polymorphism was observedin the electrophorgramof isozymes.Those species-specific EST bands detected in kidney and spleencould serve as the biochemical genetic markers of distinguishing S.meridionalis,S.asotusand their hybrid.
