低能离子注入凤仙花后DNA变异的RAPD分析

摘    要:将3种不同剂量的低能P+和N+注入来自同一纯系品种的风仙花(Impatiens balsamina L.)种子中后,采用RAID技术研究风仙花的变异。首先,建立了凤仙花的RAPD反应体系,然后,从25个随机引物中筛选出扩增条带清晰、反应结果稳定、重复性较好的3个引物,共扩增出86条较清晰的谱带,其中多态性谱带23条,分子量在350-2000bp之间。通过分析DNA的变异情况发现,注入低能离子可以使凤仙花发生DNA水平上的变异。这一研究为花卉产业的发展提供了一种新的育种手段。

Abstract:Low energy P+ and N+ ions were implanted to Impatiens balsamina L. seeds, and the best RAPD (random amplified polymorphic DNA) reaction system for Impatiens balsamina L. was established. By using random primers, DNA variation was analyzed. The results show that 3 primers were the most efficient among 25 primers, and 23 polymorphic bands were found among 86 bands gained by RAPD. The results suggest that the variation of 1mpatiens balsamina could be caused by the injection of low energy ions, and it was a useful way in flower industry.
