含CSFV T细胞表位的猪细小病毒样颗粒的表达及小鼠免疫试验

摘    要:将纯化的重组子pM-VP2-E290阳性质粒利用杆状病毒表达系统进行表达,表达产物应用SDS-PAGE、Western blot和Dot ELISA进行分析,确定所表达的蛋白分子量大小约为67kD,且具有天然蛋白的抗原特异性。应用免疫电镜对表达产物进行观察,可见到细小病毒样颗粒。病毒样颗粒在无免疫佐剂参与的情况下,免疫6~8周龄的BALB/c小鼠,同时设猪细小病毒(Porcine parvovirus,PPV)的灭活疫苗免疫组及磷酸盐缓冲液(PBS)接种组作为参比对照,检测小鼠的细胞免疫和体液免疫学指标。结果表明,表达蛋白所形成的细小病毒样颗粒,不仅能诱导小鼠机体产生抗猪瘟病毒的特异性细胞毒性T淋巴细胞(CTL)反应,还能刺激小鼠产生高效价的抗猪细小病毒的特异性抗体,而且机体所产生的抗体效价显著高于灭活疫苗对照组。

Abstract:To develop a strategy for promoting efficient antiviral immunity,the chimeric Porcine parvovirus-like particles(PPV:VLP)carrying a T cell epitope from the Classical swine fever virus(CSFV)non-structural protein NS2-3 were expressed by the baculovirus expression system and its immunogenicity on mice was analysed.SDS-PAGE,Western blot and Dot ELISA analysis showed that the expressed product was 67 kD in size and had a clear positive reaction with specific antisera against PPV and CSFV peptide.Electron microscopy analysis confirmed the particle structure of the protein.Mice immunized with these combinant particles,in the absence of adjuvant,developed high levels of PPV-specific IgG in their serum.The levels of antibodies were significantly higher than those of the antibodies from mice immunized with PPV sterile vaccine.Moreover,immunization with parvovirus-like particles carrying the CSFV T-cell epitope also elicited a peptide-specific cytotoxic-T-cell(CTL)response.These results highlight that PPV:VLP as a safe,nonreplicating antigen to develop paticulcate vaccine has high potential.
