3,5-二硝基水杨酸比色法快速测定烟草水溶性总糖、还原糖及淀粉的探讨

摘    要: 通过还原糖对3,5-二硝基水杨酸的还原产物的吸收光谱分析,结果表明,该还原产物在500 nm波长下有最大吸收峰,但最佳的工作波长确定为540 nm。同时找出了显色反应时间及显色剂的最佳用量。并对烟草中水溶性总糖的提取方法进行了优化,建立了批量连续测定烟样中还原糖、水溶性总糖(进行酸水解)、淀粉的快速方法。同时对多个烟样进行了测定,与伯川法测定结果比较,其相对误差在5%以内。

Abstract:The absorption spectrum of 3,5-dinitrosalicylic acid reducing product with reducing sugar has been studied.The result shows a maximum absorption at 500 nm,but the best work wavelength was determined by 540 nm.The best time of color reaction and best suitable amount of the color reagent was given.The extraction methods of water soluble total sugar was optimized,and a rapid continuous batch determination of reducing sugar,water soluble total sugar and starch was established.When some tobacco samples were determined.The relative errors of the results obtained by this method related to those obtained by the Bertrand's method were under of 5%.
