摘    要:开展桑树核心种质的研究有利于高效保存和利用桑树种质资源。根据15个ISSR引物扩增的98个ISSR分子标记的聚类结果和树状图,对来自山东、河北省区的46份鲁桑地方品种资源采用逐步聚类随机取样法选择核心种质,比较了样本数不同的4个核心样本群的多态性位点数、多态性位点百分率、观测等位基因数、有效等位基因数、Nei s遗传多样性指数和Shannon s信息指数等参数,最终选择了由11个样本组成的样本群作为核心种质。用统计软件SPSS对初始种质和核心种质群体的观测等位基因数、有效等位基因数、Nei s遗传多样性指数和Shannon s信息指数分别作t检验,可以看出核心种质虽然仅保留了初始种质23.91%的样品,但上述各参数在初始种质中的保有率分别为89.02%、89.03%、95%、102.24%、103.99%、101.26%,表明构建的核心种质能够代表初始种质。

Abstract:In order to conserve and utilize mulberry germplasm resources more efficiently,we developed the core collection of 46 Morus multicaulis Perr.germplasm resources from Shandong and Hebei in this paper.By using 15 selected ISSR primers,98 DNA bands were amplified from them.The result of 98 ISSR markers and the dendrogram of clustering,stepwise clustering and random methods were used to screen the core collection.Based on the parameters of 4 sample groups,including the number of polymorphic loci,percentage of polymorphic loci,and the number of alleles observed,effective number of alleles,Nei's gene diversity and Shannon's information index,11 samples were chosen as core collection.The parameters between initial collection and core collection were T-tested by using SPSS software.The results showed that the number of core collection samples is only 23.91% of initial collection,but the parameters above-mentioned of the core collection reached 89.02%,89.03%,95%,102.24%,103.99%,and 101.26% of initial collection,respectively.Those results indicated that the core collection could represent initial collection remarkably.
